Original Abstract
The fatty acid profile of fats is most frequently determined by gas chromatography of the methyl esters which are prepared by a variety of methods. Esterification increases the volatility of fatty acids, improves the peak configuration, the separation, and the sensitivity of detectors. The methyl derivatives of fatty acids are most popular because of the broad range of available methylation and transesterification procedures, suitable equipment, descriptions of derivatization procedures, and not least the long tradition of the techniques. On the other hand, esterification process is very time consuming, and some problems can occur during this procedure: incomplete conversion of fats to fatty acid methyl esters, the risk of decomposition of polyunsaturated fatty acids, formation of artefacts, loss of volatile short-chain fatty acids and others. Analysis of fatty acids in underivatized form saves the time, costs and eliminates potential errors involved in sample methylation. The aim of this work was to test and verify a simple method for the direct analysis of fatty acids by GC without derivatization, suitable for rapid analysis of fats in various food samples.
