In recent years, there has been growing interest in developing next-generation bioinoculants to contribute to sustainable agriculture. One approach to producing next-generation bioinoculants is the encapsulation Plant Growth Promoting Rhizobacteria (PGPR) through the gelation of their extracellular polysaccharides (sodium alginate). A common method for the gelation of sodium alginate involves using multivalent cations as calcium cations (Ca2+). However, the gelation of sodium alginates depends on three critical structural parameters of the polymer: molecular weight, (de)acetylation, and monomer composition. The main objective of this work is to determine the composition of the monomers. For the study of the monomer composition of sodium alginate, which is made up of L-guluronic acid blocks (poly-G) and D-mannuronic acid blocks (poly-M), the methods of ATR-FTIR, Raman microspectroscopy, and FT-Raman. The results were analyzed using principal component analysis (PCA), identifying two significant peaks for poly-M and poly-G. A calibration curve was constructed to show the relationship between the absorbance ratio of significant peaks and the percentage concentration of poly-G in the samples.
