Original Abstract
Supported lipid bilayers were used as simplified models of cell membranes to study the effect of hyaluronan solution on their diffusion properties. The bilayers were prepared from lecithin liposomes in phosphate buffer using the vesicle fusion method. Hydrophilized cover glass slides were chosen as the supporting substrate. Two fluorescence probes – Nile Red and DiO were utilized to quantify the diffusion properties of the bilayer with and without presence of hyaluronan. For this purpose Z-scan Fluorescence Correlation Spectroscopy, which is a method developed specially for planar samples, was used. It was found out, that presence of this biopolymer causes slowing of the fluorescence probe diffusion within the bilayer.
