Original Abstract
The presence of butyric acid bacteria and spores in milk causes problems in cheese technology. Some species of genus Clostridium provoke the late-blowing effect which is characterized by high concentration of butyric acid and formation of gas holes in cheese. This work is focused on application of PCR methods for genus and species identification of selected strains of Clostridium. Using genus-specific PCR, the set of 44 DNA samples was identified as DNA of Clostridium genus. Based on species-specific PCRs, it was found out that 7 samples belong to the species Clostridium butyricum and 12 samples belong to the species Clostridium tyrobutyricum. Fifteen samples were positively detected using both species-specific PCRs and therefore they were identified as DNA mixtures from both cultures. Another 10 samples were not classified into tested species. Strains with the hydrogenase (hydA) gene encoding H2-evolving hydrogenase were also searched using PCR. Specific PCR products were detected in 30 strains of the analysed set. All positive strains were identified as species Clostridium butyricum.
