The concept of next generation industrial biotechnology (NGIB) brings a possibility to obtain products of metabolism from extremophilic as same as selected mesophilic microorganisms using requirements on low sterility of whole process considering specific conditions, which predominantly extremophilic microorganisms require for their growth e.g. elevated temperature, salinity, etc. Polyhydroxyalkanoates (PHAs) represent one of the possible products which can be isolated from the cells of microorganisms living in normal as same as in extreme conditions. They offer a suitable alternative of traditional petrochemical plastics based on petroleum, whereas besides suitable physico-chemical properties are also biodegradable, biocompatible and can be advantageously produced from renewable resources. Microorganisms produce PHA in the form of hydrophobic inclusions serving as a storage of carbon, energy and necessary reducing power for biochemical processes, moreover it can protect them against different stress conditions. Therefore, for extremophiles PHA can represent one of possible adaptive mechanisms against extreme conditions of the environment. Industrially used method of isolation of PHA is based primarily on extraction of the polymer using halogenated organic solvents (predominantly chloroform) with the possibility of further purification, what it very effective, however, not very environmentally-friendly method. Use of extremophiles within NGIB concept brings an opportunity of specific approach to isolation, for instance changes in salinity of the environment (hypotonic shock) or changes of temperature together with detergents for enhancement of purity of isolated material. By combination of the parameters we can gain an more ecological isolation of PHA of sufficient purity for targeted applications.
