Original Abstract
The treatment of chronic wounds is currently a global problem, not only in terms of time and money, but also due to the increase in bacterial resistance to antibiotics. As a suitable replacement for failing antibiotics, phage therapy is currently offered, where the bacteriophage (virus) attacks exclusively bacterial cells, and multiplies spontaneously at the site of infection. In combination with a lyophilized hydrogel film based on natural Gum Karaya polysaccharide (GK), which is able to provide a moist environment to support wound healing and visualization, we obtain a multifunctional material with the potential to accelerate the treatment of chronic wounds. The sorption capacity of the hydrogel carrier holding up to 300% of the volume of water compared to its original weight, is important in terms of transparency and ensuring a moist wound environment, while being stable for 28 days. Sufficient mechanical properties were confirmed by tensile strength, when the material elongated by up to 140% in the wet state. The gradual release of more than 90% of bacteriophages from the hydrogel structure was monitored by the bilayer agar method. Antimicrobial activity of phage and hydrogel tested on a strain of methicillin-resistant Staphylococcus aureus ATCC 43300 (MRSA) demonstrated positive phage activity. Physicochemical properties of GK film indicated its potential as a suitable hydrogel carrier for use in long-term treatment of chronic wounds, thus reducing the need for frequent replication. In addition, antibacterial assays have confirmed that GK hydrogel films sorbed by bacteriophages are effective on MRSA, thus potentially shortening the healing time of a chronic defect.
