Steady-state fluorescence techniques represent a powerful tool for the determination of great variety of phospholipids vesicles (liposome) properties. Phase transition temperature is one of the most important characteristics of liposomes. It is commonly determined by means of calorimetric technics. It can be also obtained by analyzing of laurdan emission spectra at different temperatures. The degree of a lipid acyl chain order in liposomes and a membrane fluidity can by studied by means of fluorescence anisotropy, which measures depolarization of fluorophores excited by polarized light, where the depolarization depends on the membrane fluidity. Diphenylhexatriene is a suitable fluorescence probe for the anisotropy measurements. The aggregation behavior of lipid molecules can be studied using fluorescence probes such as pyrene or perylene. These fluorescence probes are sensitive to the polarity of the local environment and indicate changes in lipid aggregates character. Fluorescent lipophilic tracers such a DiO or DiA are usually applied in a staining of the cell surface, but they can be easily used also in aggregates formation study.
