DETERMINATION OF ANTIBIOTICS IN WASTEWATER

Antibiotic are used in human and veterinary medicines to treat diseases. These compounds have been shown to block biological processes in wastwater treatment plants. They harm natural ecosystems and may concentrate in food chain. Furthermore, the bacterial resistance against them has been proven to establish easily. Sulfonamides are derivates of p-aminobenzenesulfonamide. This group of antibiotics is widely used as antibacterial drugs in medical and veterinary applications. They are often prescribed for treatment of many human and animal infectious diseases. The analysis of sulfonamides is influenced by their properties, namely insolubility in water, ethanole, ether and dichloromethane. The samples were collected from the municipal wastewater treatment plant and the veterinary wastewater treatment plant in Brno, The Czech Republic. The simple extraction for sulfonamides (sulfathiazole) in wastewater was developed. Sulfathiazole was extracted by liquid-liquid extraction with ethylacetate and evaporated by a rotary vaccuum evaporator. The evaporation residues were dissolved in methanole and mobile phase. The mixture of acetonitrile and 0,1% formic acid in purified water was used as the mobile phase. The samples were filtered by membrane filters (0,45 m) into the amber glass vials. The HPLC column used for determination was Zorbax Eclipse XDB C18 (4,6 x 150 mm, 5 m) and elution was carried out using gradient mode. The HPLC instrument equipped with diode array detector was used for antibiotic determination.
