In our study, we used bacterial strain Rhodospirillum rubrum (ATCC 11170), a purple, non-sulfur, Gram-negative bacteria, which has extraordinary metabolic versatility and adaptability as it was showed to be able to grow both aerobically and anaerobically, both under hetero- and autotrophic conditions, and is a facultative phototroph (anaerobic prerequisites) both in heterotrophic and autotrophic conditions. Furthermore, this bacteria is also capable of production of pigments (carotenoids) in specific conditions (anaerobic) and fixation and utilization of CO and CO2. Therefore, it this strain could be used for valorization of waste gases such as combustion products or syngas. In this work, we focused on the influence of aeration on growth of bacterial strain and its production of PHA in dark/light environment. Several groups of instruments were used, firstly we characterized optical densities of individual cultures and CFU (UV-Vis spectrophotometer, flow cytometer). We used also microscopic techniques such as Cryo-SEM and FLIM for characterization of morphology of intracellular space and surface of cells. Last technique which were used was UV-Vis absorption spectroscopy in diffuse reflection mode to investigate and correlate amounts of carotenoids with obtained data from extraction experiments.
